class=\"wp-block-heading\" id=\"\u7814\u7a76\u65b9\u5411\">\u7814\u7a76\u65b9\u5411<\/h1>\n\n\n\n<p class=\"has-medium-font-size\">Antibody producing B cells arise from hematopoietic stem cells through a series of developmental steps that involve a hierarchical transcription factor network. Whereas much is known about their direct genetic targets, little is known about their actual mechanism of action through interactions with various coactivators that act on chromatin structure or on the general transcription machinery. In addition, genetic mutations of the involved transcription factors result in loss-of-function or gain-of-function frequently lead to B cell deficiency or malignancy.<\/p>\n\n\n\n<p class=\"has-medium-font-size\">Our goal is to uncover the mechanisms by which transcriptional activators\/coactivators (including various histone modifiers\/remodelers) control the development and leukemogenesis of B-lymphocytes, as well as searching therapeutic targets\/approaches for B-cells associated leukemia\/lymphoma.<br>On-going studies will employ various state-of-the-art biomedical technologies that include:<\/p>\n\n\n\n<ol class=\"wp-block-list has-medium-font-size\">\n<li>biochemically isolating recombinant proteins\/ protein complexes from bacteria\/insect\/mammalian cell cultures for cell-free assays<\/li>\n\n\n\n<li>genome-widely profiling (ChIP-seq and RNA-seq) the transcription-related factors (DNA-binding proteins and transcriptional coactivators) and corresponding histone modifications in normal or malignant cells<\/li>\n\n\n\n<li>developing the peptidomimetics for potential therapeutic to leukemia\/lymphoma treatment<\/li>\n<\/ol>\n\n\n\n<p class=\"has-medium-font-size\">\u57fa\u56e0\u8f49\u9304\u662f\u7d30\u80de\u5c55\u73fe\u751f\u547d\u73fe\u8c61\u7684\u91cd\u8981\u74b0\u7bc0\u4e4b\u4e00\uff0c\u800c\u4e0d\u6b63\u5e38 \u7684\u57fa\u56e0\u8f49\u9304\u4e5f\u662f\u9020\u6210\u75be\u75c5\u8207\u764c\u75c7\u7684\u4e3b\u8981\u539f\u56e0\uff0c\u6240\u4ee5\u7814\u7a76\u57fa\u56e0\u8f49\u9304\u7684\u5206\u5b50\u6a5f\u8f49\u662f\u751f\u547d\u79d1\u5b78\u4e2d\u4e00\u500b\u57fa\u672c\u4e14\u91cd\u8981\u7684\u8ab2\u984c\u3002\u904e\u53bb 40 \u5e74\u7684\u7814\u7a76\u4e2d\uff0c\u9019\u500b\u9818\u57df\u5df2\u7d93\u767c\u73fe\u81f3\u5c11\u9700\u8981 100 \u591a\u7a2e\u86cb\u767d\u7684\u53c3\u8207\u624d\u80fd\u8f49\u9304 DNA \u4e0a\u7684\u57fa\u56e0\u8cc7\u8a0a\uff0c\u4f46\u662f\u5c0d\u65bc\u9019\u4e9b\u8f49\u9304\u76f8\u95dc\u86cb\u767d\u9593\u7684\u5404\u7a2e\u4ea4\u4e92\u4f5c\u7528\u5247\u6240\u77e5\u751a\u5c11; \u53e6\u5916\u9ad8\u901a\u91cf\u7684\u6b21\u4e16\u4ee3\u5b9a\u5e8f\u6280\u8853\u4e5f\u5c07\u57fa\u56e0\u8f49\u9304\u7684\u7814\u7a76\u5f9e\u4ee5\u5f80\u7684\u55ae\u4e00\u57fa\u56e0\u63a8\u5c55\u5230\u5168\u57fa\u56e0\u9ad4\u7684\u5c64\u7d1a\uff0c\u9019\u500b\u6280\u8853\u767c\u5c55\u5e6b\u52a9\u77ad\u89e3\u8f49\u9304\u4f5c\u7528\u7684\u666e\u904d\u6cd5\u5247\uff0c\u66f4\u91cd\u8981\u7684\u662f\u80fd\u63d0\u4f9b\u7cbe\u6e96\u91ab\u5b78\u7684\u91cd\u8981\u4f9d\u64da\u3002<\/p>\n\n\n\n<p class=\"has-medium-font-size\">\u76ee\u524d\uff0c\u5be6\u9a57\u5ba4\u7684\u7814\u7a76\u4e3b\u984c\u4e4b\u4e00\u662f\u60f3\u77ad\u89e3\u8f49\u9304\u56e0\u5b50 E2A \u878d\u5408\u86cb\u767d\u9020\u6210 B \u7d30\u80de\u816b\u7624\u7684\u5206\u5b50\u6a5f\u8f49\uff0c\u4ee5\u53ca\u6240\u8abf\u63a7\u7684\u57fa\u56e0\u7d44\u3002 \u6211\u5011\u767c\u73fe E2A \u878d\u5408\u86cb\u767d\u80fd\u626e\u6f14\u4e00\u822c\u8f49\u9304\u8f14\u56e0\u5b50\u7684\u529f\u80fd\uff0c\u4fc3\u9032\u53e6\u4e00\u500b\u8f49\u9304\u56e0\u5b50 RUNX1 \u7684\u6d3b\u6027\u800c\u9020\u6210\u8840\u7403\u5e79\u7d30\u80de\u7121\u6cd5\u5206\u5316\uff0c\u9019\u500b\u767c\u73fe\u5728\u672a\u4f86\u80fd\u63d0\u4f9b\u6cbb\u7642B\u7d30\u80de\u816b\u7624\u7684\u7cbe\u6e96\u91ab\u7642\u7b56\u7565\u3002
