(autophagy)\u70ba\u5e6b\u52a9\u7d30\u80de\u5728\u4e0d\u5229\u74b0\u5883\u4e0b\u5b58\u6d3b\u7684\u91cd\u8981\u53cd\u61c9\u3002\u7c92\u7dda\u9ad4\u81ea\u566c\uff08mitophagy\uff09\u662f\u4e00\u7a2e\u7279\u5b9a\u5f62\u5f0f\u7684\u81ea\u566c\u4f5c\u7528\uff0c\u91dd\u5c0d\u7c92\u7dda\u9ad4\u9032\u884c\u54c1\u8cea\u7ba1\u63a7\u3002\u901a\u904e\u6b64\u6a5f\u5236\uff0c\u771f\u6838\u7d30\u80de\u80fd\u5920\u9078\u64c7\u6027\u6e05\u9664\u591a\u9918\u6216\u640d\u58de\u7684\u7c92\u7dda\u9ad4\u4f86\u7dad\u6301\u7d30\u80de\u5065\u5eb7\u3002\u7576\u7c92\u7dda\u9ad4\u640d\u50b7\u6642\uff0c\u7c92\u7dda\u9ad4\u819c\u96fb\u4f4d\u7684\u4e0b\u964d\u6703\u5c0e\u81f4PINK1\uff08\u4e00\u7a2e\u7d72\u6c28\u9178\/\u8607\u6c28\u9178\u6fc0\u9176\uff09\u5728\u7c92\u7dda\u9ad4\u5916\u819c\uff08OMM\uff09\u4e0a\u7684\u7d2f\u7a4d\u3002\u7a69\u5b9a\u7684PINK1\u6703\u78f7\u9178\u5316\u8207\u7c92\u7dda\u9ad4\u5916\u819c\u86cb\u767d\u4e0a\u7684\u6cdb\u7d20\uff08ubiquitin\uff09\uff0c\u5f9e\u800c\u4f7fParkin\uff08\u4e00\u7a2e\u6cdb\u7d20E3\u9023\u63a5\u9176\uff09\u805a\u96c6\u5230\u640d\u58de\u7684\u7c92\u7dda\u9ad4\uff0cParkin\u5728\u5176\u4e0a\u5c0d\u591a\u500b\u5916\u819c\u86cb\u767d\u9032\u884c\u591a\u6cdb\u7d20\u5316\uff08polyubiquitination\uff09\u3002\u6700\u7d42\uff0c\u640d\u58de\u7684\u7c92\u7dda\u9ad4\u6703\u88ab\u6cdb\u7d20\u7d50\u5408\u7684\u81ea\u566c\u53d7\u9ad4\uff08autophagy receptors\uff09\u8b58\u5225\uff0c\u4e26\u4fc3\u9032\u7c92\u7dda\u9ad4\u81ea\u566c\u4f5c\u7528\u7684\u9032\u884c\u3002<\/p>\n\n\n\n<p class=\"has-medium-font-size\">Macroautophagy (hereafter referred to as autophagy) is a catabolic pathway by which cells sequester unwanted or damaged cellular proteins or organelles through a double membrane structure called the autophagosome. This process is mediated by a set of evolutionarily conserved genes, the autophagy-related (ATG) genes, which function in nucleation of the autophagosomal membrane, elongation of the autophagic membrane, sequestration of cytoplasmic constituents, and lysosomal degradation of the sequestered contents.<\/p>\n\n\n\n<p class=\"has-medium-font-size\">Selective autophagy is a homeostatic quality control process that targets specific cytoplasmic components to autophagosomes for lysosomal destruction. Diverse cargos have been identified as substrates for selective autophagy, including mitochondria (mitophagy), peroxisomes (pexophagy), endoplasmic reticulum (ERophagy), and viruses (virophagy). Mitophagy is the major pathway by which eukaryotic cells eliminate damaged or unwanted mitochondria as damaged mitochondria release reactive oxygen species (ROS), leading to inflammasome activation, genotoxic stress, promotion of tumorigenesis and aging. Defects in mitophagy are implicated in neurodegenerative diseases such as Parkinson\u2019s disease, Alzheimer disease, and age-related pathologies. A fundamental question in the autophagy field remains how cargos such as mitochondria are selectively targeted for autophagic degradation. We will use series of genetic, cell biology, biochemical approaches to understand<strong> the mechanisms that confer substrate selectivity<\/strong> in mitophagy and investigate the <strong>pathological consequences of mitophagy dysfunction in diseases<\/strong> using model organisms such as C. elegans and mice.
